Overview
Retatrutide is an investigational peptide that activates three hormone receptors involved in energy balance: GIP, GLP-1 and glucagon. Adding glucagon-receptor activity to the incretin pair is thought to increase energy expenditure on top of the appetite effects of GLP-1.
It is still in clinical development. A phase 2 obesity trial published in 2023 reported the largest average weight reduction seen with an incretin-based peptide at that time, and phase 3 trials are ongoing.
Research areas
- Body-weight regulation and energy expenditure
- Glycaemic control in type 2 diabetes models and trials
- Liver fat and metabolic dysfunction-associated steatotic liver disease
- Contribution of glucagon-receptor signalling to multi-agonist effects
